As a digestive protease, pepsin originates from gastric chief cells of stomach lining. Cells secrete pepsinogen, which matures into active pepsin when exposed to gastric acid or pepsin. It digests food proteins into short peptides, and some peptides are split into amino acids such as tyrosine and phenylalanine.

UV spectrophotometry: Under test conditions (37±0.5°C), one unit (u/g) refers to the amount of enzyme that hydrolyzes hemoglobin to produce trichloroacetic acid-soluble substances with absorbance equivalent to 1 μmol tyrosine at 275 nm per minute.
